Abstract

Die DNA-Methylierung spielt in der epigenetischen Vererbung und in der Entstehung von Krebs eine wichtige Rolle. Vor kurzem wurde gezeigt, dass die DNA-Methylierung in Drosophila Embryonen von Dnmt2 vermittelt wird. Dnmt2-Enzyme besitzen im Gegensatz zu anderen eukaryotischen DNA-Methyltransferasen keine ausgedehnte N-terminale „regulatorische“ Domäne, sondern bestehen vielmehr aus einer kompakten katalytischen Domäne mit hoch konservierten DNA-Methyltransferase-Motiven. Die Mechanismen, die der Regulation und Zielsteuerung dieser Enzyme zu Grunde liegen, sind noch unbekannt. Deshalb wurde die Möglichkeit in Betracht gezogen, dass Dnmt2 durch Faktoren reguliert sein könnte, welche die fehlende „regulatorische“ Domäne durch Protein-Protein-Interaktionen ersetzen. Da die höchsten Mengen an 5-methyl-Cytosin während der frühen Embryogenese detektiert wurden, erfolgte zur Identifizierung von Dnmt2-interagierenden Proteinen ein Yeast Two-Hybrid Screen mit einer embryonalen Drosophila cDNA-Bibliothek. Als primärer Interaktionspartner von Dnmt2 wurde dabei ein bisher uncharakterisiertes Protein identifiziert, das im Folgenden als „IPOD“ (Interaction Partner Of Dnmt2) bezeichnet wird. Die Interaktion zwischen Dnmt2 und IPOD konnte in Co-Immunopräzipitations-Assays und GST Pull-down Assays bestätigt werden. Dabei gelang der Nachweis, dass die N-terminale Domäne von Dnmt2 für die Wechselwirkung mit IPOD verantwortlich ist. In Übereinstimmung mit der Konservierung von Dnmt2 in Drosophiliden zeigte eine nähere Sequenzanalyse, dass das IPOD-Protein in diesen Spezies hoch konserviert ist. Des Weiteren wies eine Western Blot Analyse mit einem spezifisch gegen IPOD etablierten Antiserum auf eine entwicklungsspezifische Expression von IPOD während der frühen Embryogenese hin. Eine Assoziation von IPOD mit chromosomaler DNA und eine Co-Lokalisierung von Dnmt2 mit IPOD konnte Zellzyklus-abhängig sowohl in Drosophila Embryonen als auch spezifisch in Polytän-Chromosomen aus Speicheldrüsen IPOD-überexprimierender Larven nachgewiesen werden. In Übereinstimmung mit der Co-Lokalisierung beider Proteine in Metaphasen führte die RNAi-vermittelte Reduktion von IPOD in Drosophila Embryonen zu einem Verlust der Assoziation von Dnmt2 an Metaphase-Chromosomen. Darüber hinaus resultierte die Überexpression von IPOD durch ein induzierbares Transgen in Entwicklungsstadien von Drosophila melanogaster, in denen endogen keine DNA-Methylierung nachweisbar ist, in eine Induktion der Dnmt2-Expression und in eine signifikante DNA-Hypermethylierung des Fliegen-Genoms. Diese Ergebnisse legen nahe, dass die DNA-Methylierung in Drosophila durch die Interaktion von Dnmt2 mit IPOD reguliert wird.

Translation of abstract (English)

DNA methylation plays an important role in epigenetic inheritance and cancer. It has been recently shown that DNA methylation in Drosophila embryos is mediated by Dnmt2. Dnmt2 enzymes contain only a catalytic DNA methyltransferase domain, while in contrast to other eukaryotic DNA methyltransferases, they lack an extended N-terminal “regulatory“ domain. The mechanisms underlying the regulation and targeting of these enzymes are unknown. The possibility that Dnmt2 might be regulated by accessory factors that substitute for the regulatory domain via protein-protein interactions was addressed. Based on the finding that the highest levels of 5-methylcytosine occured during early embryonic stages, a yeast two-hybrid screen with an embryonic Drosophila cDNA library was performed to identify Dnmt2-interacting proteins. A previously uncharacterized protein, “IPOD“ (Interaction Partner Of Dnmt2), was identified that showed strong and consistent interaction with Dnmt2. The interaction between Dnmt2 and IPOD was confirmed via co-immunoprecipitation and GST pull-down assays. The region of Dnmt2 responsible for the interaction was mapped to the N-terminal domain. Consistent with the conservation of Dnmt2 in Drosophilidae, further analysis of the protein revealed that IPOD is highly conserved in these species. Western Blot analysis by a specific antiserum against IPOD revealed developmentally regulated expression during embryogenesis. An association of IPOD with chromosomal DNA and a co-localization of Dnmt2 and IPOD was detected in a cell-cycle dependent manner during metaphases in Drosophila embryos and in salivary gland polytene chromosomes of IPOD-overexpressing larvae. Furthermore, consistent with our hypothesis, the RNAi-mediated knockdown of IPOD in Drosophila embryos resulted in a loss of Dnmt2-association to metaphase chromosomes. In addition, overexpression of IPOD from an inducible transgene during stages of Drosophila development, where no endogenous DNA methylation is detectable, induced the expression of Dnmt2 and resulted in significant hypermethylation of the fly genome. These results suggested that DNA methylation in Drosophila is regulated by the interaction betwenn Dnmt2 and IPOD.
