Abstract

New receptor-based models for pattern formation and regulation in multicellular biological systems are proposed. The test organism for the mathematical modelling is a fresh-water polyp, hydra. Another model is applied to describe the growth of tumour along linear or tubular structure. Models are defined in the form of reaction-diffusion equations with zero flux boundary conditions coupled with ordinary differential equations. Two mechanisms of pattern formation: diffusion driven instability and hysteresis-driven mechanism are studied and their possibilities and constraints in explanation of different aspects of pattern formation and regulation are demonstrated. Three-variable (describing the dynamics of ligands, free and bound receptors) and four-variable models (including also an enzyme cleaving the ligand) are analysed and compared. It is shown under which conditions they can involve diffusion driven instability mechanism for pattern formation. It is shown that gradient in the density of bound receptors occurs if there is also a second diffusible substance, an enzyme, which degrades ligands. The four-variable model is able to capture results from cutting experiments and reflects {\it{de novo}} pattern formation from dissociated cells. To explain the grafting experiments a new model including memory-based relation is proposed. Production of the diffusible biochemical molecules has a hysteretic dependence on the density of these molecules and is modelled by additional ordinary differential equations. The stationary and oscillatory patterns, resulting from multiple steady states and switches in the production rates, are found. Finally, a receptor-based approach is applied to a model of the growth of a tumour. Mathematical model describing the population of cells distributed over a linear or tubular structure and the diffusing growth factor which regulates the proliferation is derived and analysed.

Translation of abstract (German)

In dieser Arbeit werden neue Modelle vorgestellt, die mögliche Musterbildung in vielzelligen biologischen Systemen und ihre Steuerung beschreiben. Als Vergleichsorganismus dient der Süßwasserpolyp Hydra. Ein weiteres Modell beschreibt das Tumorwachstum entlang von geraden oder röhrenförmigen Strukturen. Diese Modelle sind in Form von Reaktions-Diffusions-Gleichungen ohne Fluss durch den Rand dargestellt sind mit gewöhnlichen Differentialgleichungen gekoppelt. Für die Musterbildung werden zwei Zusammenhänge untersucht: Die durch Diffusion hervorgerufene Instabilität und die Hysterese. Ihre Möglichkeiten aber auch ihre Unzulänglichkeiten für die Erklärung der verschiedenen Aspekte der Musterbildung und ihre Regelung werden dargestellt. Es werden Modelle untersucht, die die Dynamik der drei Variablen: Zahl der freien Liganden, der freien Rezeptoren und der Ligand-Rezeptorkomplexe beschreiben. Es wird gezeigt, unter welchen Bedingungen diese zu Muster- bildung führen, die durch Diffusion-bedingte Instabilität hervorgerufen wird. Es wird gezeigt, daß ein Gefälle in der Dichte der gebundenen Rezeptoren auftaucht, falls eine weitere diffusionsfähige Substanz hinzukommt, hier ein Enzym, welches die Liganden abbaut. Das Modell mit vier Variablen ist in der Lage die Ergebnisse von Versuchen darzustellen, in denen die Körpersäule quer geteilt wurde, und die Wiederherstellung je eines vollständigen Körpermusters in beiden getrennten Körperhälften vorherzusagen. Um Ergebnisse von Transplantationsexperimenten erklären zu können wird ein neues Modell mit Gedächtnis vorgeschlagen. Die Produktionrate des diffusionsh"abhängigen biochemischen Moleküls (Liganden) hängt Hysterese-artig vom Anteil dieser Moleküle ab und wird durch weitere gewöhnliche Differentialgleichungen modelliert. Stationäre und oszillierende Muster, die von mehrfachen Ruhezuständen und einem Umschalten der Produktionsraten hervorgerufen werden, konnten erzeugt werden. Zum Abschluß wird der Formalismus des auf Rezeptoren gegründeten - Modells für die Beschreibung von Tumorwachstums angewandt. Diese führt zu einem mathematischen Modell, welches zum einen die Verteilung der Zellen, die auf eine gerade bzw. röhrenförmige Struktur verteilt sind, und zum anderem den diffundierenden Wachstumsfaktor beschreibt, der die Proliferation steuert.
