Abstract

Ziel dieser Studie war den Beitrag von Semaphorin 4D und Plexinen der Gruppe B bei der Entwicklung der Zytoarchitektur von hippocampalen Neuronen näher zu untersuchen. Bisher wird der Sema4D-Plexin-B1 Wechselwirkung im zentralen Nervensystem eine klassische Rolle bei den Semaphorinwirkungen zugeordnet: der Kollaps von axonalen Wachstumskegeln. Dieses Phänomen ist bei hippocampalen und retinalen Neuronen während einer frühen Phase der Neuritenentwicklung aufgezeigt worden. Über die funktionelle Bedeutung der Sema4D-Plexin-B Wechselwirkung bei den nachfolgenden Entwicklungsphasen von Neuriten, der Differenzierung und Reifung, existieren zum Zeitpunkt dieser Arbeit keine Untersuchungen. Die vorliegende Dissertation beschäftigte sich mit dieser Fragestellung zum einen durch Video-Mikroskopiebeobachtungen und Untersuchungen zur Morphologie hippocampaler Neuronen und zum anderen durch die Analyse hippocampaler Pyramidalneuronen von Plexin-B1 und Plexin-B2 Knockout-Mäusen. Die Behandlung embryonaler hippocampaler Neurone mit Sema4D führte innerhalb von 4 Stunden zu einem Anstieg der dendritischen Verzweigungen und der neuronalen Komplexität. Ferner konnte anhand von Inhibitorexperimenten nachgewiesen werden, dass die Effekte auf die Dendriten über die Rezeptortyrosinkinase Met, PI3K und mTOR vermittelt wurden. Die axonale Verzweigung hing hingegen von der koordinierten Aktivierung der GTPase RhoA und den beiden Rezeptortyrosinkinasen Met und ErbB-2 ab. Über Experimente mit hippocampalen Neuronen aus Plexin-B1 Knockout Mäusen und der Überexpression von dominant-negativem Plexin-B2 konnte nachgewiesen werden, dass sowohl Plexin-B1, als auch Plexin-B2 an der Signaltransduktion beteiligt waren. Überraschenderweise zeigten sowohl adulte Plexin-B1 Knockout-Mäuse, als auch am postnatalen Tag 5 geborene Plexin-B1 und Plexin-B2 Knockout-Mäuse keine signifikanten Unterschiede in ihrer hippocampalen Schichtung und der Verzweigung hippocampaler Pyramidalneurone. Dies verdeutlicht, dass bei der Morphogenese, als ein essentieller Schritt der Entwicklung von Neuronen, das Wegfallen der Plexine durch weitere Faktoren kompensiert werden konnte oder die Expression der Plexin generell für die Verzweigung von Pyramidalneuronen der CA1-Region keine Bedeutung hatte. Neben den Effekten auf die Verzweigung von Axonen und Dendriten führte die Applikation von Sema4D auch zu einem Anstieg der Anzahl an Filopodien. Es handelt sich bei der vorliegenden Arbeit um die erste Studie, die eine koordinierte Aktivierung von B-Plexinen mit mTor und einem Anstieg der Verzweigung von Neuriten in direkte Verbindung stellt.

Translation of abstract (English)

The aim of this study was to assess the contribution of Semaphorin 4D and Plexin-B proteins in shaping the cytoarchitecture of the hippocampus and of developing hippocampal neurons. Until now, Sema4D-Plexin-B interactions in the central nervous system has been assigned a classical role of semaphorin actions: the collaps of axonal growth cones. This has been demonstrated in hippocampal and retinal neurons over early stages of neurite outgrowth. However the functional role of Sema4D-Plexin-B interactions over subsequent stages of neurite development, differentiation and maturation has not been characterised. In the current dissertation project, this question was addressed using morphogenetic assays and time-lapse imaging on developing hippocampal neurons a model system as well via the analysis of knockout mice lacking the Plexin-B1 or Plexin-B2 proteins. Treatment of developing hippocampal neurons with Sema4D resulted in a promotion of dendritic branching and neuronal complexity within 4 hours. Furthermore, using specific inhibitors of Plexin coreceptors and downstream targets, we observed that an activation of the receptor tyrosine kinase Met, PI3K and mTOR was crucial for Sema4D-mediated effects on dendritic development, whereas the coordinated activation of the GTPase RhoA, and the two receptor tyrosin kinases Met and ErbB-2 was mandatory for Sema4D mediated axonal branching. Experiments using Plexin-B1 knockout mice and overexpression of a dominant-negative Plexin-B2 construct revealed that both, Plexin-B1 and Plexin-B2 are required for Sema4D-mediated effects on hippocampal neurons. Surprisingly, analysis of adult Plexin-B1 knockout mice and of postnatal day 5 Plexin-B1 and Plexin-B2 knockout mice showed no significant differences in the development of hippocampal layering and branching of hippocampal pyramidal neurons, respectively. This could result from a redundant expression of Plexin-B1 and Plexin-B2 in developing pyramidal neurons or from a compensation by additional factors. Besides its effect on axonal and dendritic branching Sema4D was observed to increase the number of filopodia in hippocampal neurons. This is the first study that links activation of B-Plexins to mTor and to an functional increase in dendritic and axonal branching of hippocampal neurons.
