Abstract

Im Rahmen der Entwicklung zu schmerzfreien Methoden für die Behandlung von Diabetes beschäftigt sich diese Arbeit mit der Konzeption, dem Aufbau und der ersten Erprobung eines kompakten nicht-invasiven Blutglukose-Messgerätes für Mäuse. Zu diesem Zweck wurde die aus dem Schwanz der Maus austretende Infrarotstrahlung genutzt, die aufgrund der Differenz zwischen Körper- und Außentemperatur eine Temperaturgradientenregion innerhalb der äußeren Hautschichten durchläuft. Dieser Temperaturgradient entspricht einer Durchstrahlung der Hautschichten, wodurch ein Absorptionsspektrum der beinhalteten Substanzen erzeugt wird. Im Fingerprint-Bereich der Glukose zwischen 9-10µm wird dadurch eine Absorptionsmessung und somit eine Konzentrationsbestimmung von Glukose in den Hautschichten möglich. Die Umsetzung dieser Messung fand mit einer speziell für Mäuse entwickelten Infrarot-Messeinheit statt, mit der die Geometrie des Mäuseschwanzes optimal detektiert werden kann. Zur exakteren Bestimmung der Werte wurden mehrere Einzeldetektoren verwendet, wovon ein Referenzkanal die Hintergrundstrahlung misst. Das Rauschen der ankommenden Signale wurde zusätzlich mittels Modulation durch ein Chopperrad und einen in der Software integrierten Lock-in-Effekt stark reduziert. Bei den Testmessungen konnte beim Vergleich mit konventionellen Glukosetoleranztests gezeigt werden, dass das hier entwickelte Gerät zur Glukosekonzentration proportionale Messwerte liefern kann. Vergleichmessungen mit einem implantierten Sensor zeigten einen nahezu identischen Verlauf der Glukosewerte, wodurch die prinzipielle Funktionsfähigkeit des Messprinzips deutlich bestätigt wurde.

Translation of abstract (English)

Focussing on the development of pain-free methods for the treatment of diabetes, this thesis discusses the design, the construction and the first field tests of a new non-invasive blood- glucose-analyser for mice. For this purpose the infrared radiation from a mouse tail was used. Thermal radiation passes through a temperature gradient between the body and the ambient temperature within the outer skin layers. The temperature gradient causes an effective radiography of the colder skin layers so an absorption spectrum of the finger-print-region between 9 and 10µm is obtained allowing the determination of the glucose concentration. The realisation of these measurements reqired an optical geometry adjusted for the optimal detection of the tail radiation. The implementation of a signal modulation and a lock-in-effect within the software reduces additionally the noise of the measurement. The constructed analyser delivers signals proportional to glucose concentration. Continious glucose measurements were done and compared to an implanted sensor. The glucose values found in both methods are similar, proofing the concept for the non-invasive blood-glucose-analyser developed in the thesis.
