Abstract

In dieser Arbeit sollte das Potential synthetischer Saccharidmimetika getestet werden, verschiedene Schritte der metastatischen Kaskade in vitro zu hemmen. Dabei wurden an Melanomzellen einige ausgewählte Strukturen zweier Substanzbibliotheken auf ihre Wirkung gegenüber der Zell-Adhäsion und -Migration sowie der Aktivierung von Matrix-Metalloproteinasen untersucht. Erste Versuche mit galactosylierten Cyclohexan-Derivaten haben gezeigt, daß sich die Adhäsion von B16F10-Mausmelanom-Zellen an Fibronectin in Anwesenheit von 40 mM 1,3-Bis-beta-D-galactopyranosyl-oxymethyl-5-hydroxymethyl-cyclohexan um 80 % inhibieren läßt. Die Wirksamkeit von Galactose als aktives Element bestätigten Adhäsionsversuche mit digalactosylierten Furan-Derivaten, welche in äquivalenter Konzentration in der Lage waren die Adhäsion der B16F10-Zellen an Fibronectin (40 %), Vitronectin (50 %) und Laminin (50 %) signifikant zu reduzieren. Der Furanring als Grundgerüst der Saccharidbibliothek bietet dabei den Vorteil, daß das Saccharidmimetikum über eine Diels-Alder-Reaktion an Fluoreszenz-Farbstoffe gekoppelt werden kann. Auf diese Weise lassen sich Moleküle erzeugen, die als Marker zuckerbindender Strukturen zur Aufklärung des Wirkmechanismus der Saccharidmimetika genutzt werden können. Die Substitution eines Galactoserestes am Furan-Ring gegen eine Fucoseeinheit ergab keine Verbesserung der inhibitorischen Eigenschaften. Auch eine Übertragung der Ergebnisse auf die humane Melanom-Zellinie WM-115 ergab für 40 mM 3,4-Bis-beta-D-galactopyranosyl-oxymethyl-furan nur noch einen geringen Rückgang der Adhäsion an Fibronectin (26 %) bzw. Fibrinogen (30 % ab 5mM). Ein Effekt der Substanz auf die Aktivität der Matrix-Metalloproteinase-2 bzw. das Migrationspotential der Zellen war nicht zu erkennen. Die Addition einer geladenen Sulfatgruppe an den Furanring ergab schließlich die gewünschte Optimierung der Wirksamkeit des Saccharidmimetikums. 3-beta-D-Galactopyranosyl-oxymethyl-4-sulfatomethyl-furan (GSF) ist in der Lage die Adhäsion des B16F10 Maus-Melanoms und der humanen WM-115-Linie an Fibronectin (40 mM) und Fibrinogen (20 bzw. 10 mM) vollständig zu unterdrücken. Es verminderte die Aktivierung der MMP-2 und inhibierte die Migration von Zellen in einer Konzentration von 5 mM. Ein Rückgang der Zellzahl um 50 % aufgrund cytotoxischer Effekte ist bei dieser Konzentration erst nach mehr als 60 h erkennbar und daher für die beschriebenen Versuche nicht relevant. Die vollständige Inhibition der Adhäsion, sowie die signifikanten Auswirkungen auf die Aktivierung von MMP-2 und die Zell-Migration deuten auf eine Interaktion von GSF mit einem oder mehreren Mitgliedern der Integrinfamilie hin. Anhand der vorliegenden Arbeit ist es gelungen zu zeigen, daß insbesondere geladene Saccharidmimetika das Potential besitzen die metastasierenden Eigenschaften von Melanomzellen in vitro zu unterdrücken. Das Screening nach weiteren geladenen Saccharidmimetika auf Furanbasis, deren vergleichbare Wirkung in geringeren Konzentrationen vorhanden sein sollte, scheint erfolgsversprechend und die Substanzen sollten durch die Möglichkeit einer kombinatorischen Synthese zugänglich sein.

Translation of abstract (English)

The aim of this work is to investigate the potential of synthetic saccharide mimics to interfere with different steps of the metastatic cascade in vitro. Different structures of two libraries of saccharid mimics were tested for their capacity to inhibit cell-adhesion and –migration as well as the activation of matrix-metalloproteinases (MMPs) of melanoma cells. Treatment of the melanoma cells with galactosylated derivatives of cyclohexane had shown that cell-adhesion of murine B16F10 melanomas to fibronectin decreased about 80 % in presence of 40 mM 1,3-Bis-beta-D-galactopyranosyl-oxymethyl-5-hydroxymethyl-cyclohexane. The inhibitory effect of saccharide mimics containing galactose in adhesion assays was confirmed by using di-galactosylated derivatives of furan which reduced the adhesion of B16F10 melanomas to fibronectin by about 40 % and to vitronectin and laminin by about 50 %. The furanoic core-structure of the library shows advantages over cyclohexane, because furan can thus react in a Diels-Alder reaction with fluorescent maleimides. Diagnostic tools can be generated to investigate the interaction of the saccharide mimic with the cell surface. Chemical exchange of galactose against fucose does not improve the inhibitory effect of the molecule. The adhesion of human melanoma cell line WM-115 to fibronectin decreased by about 26 % and to fibrinogen by 30 % in the presence of 40 mM 4-Bis-beta-D-galactopyranosyl-oxymethyl-furan. An effect of the saccharide mimic on the activation of MMP-2 or upon cell-migration was not observed. Introduction of a sufate-residue has led to a significant improvement of the inhibitory effect of the saccaride mimic. 3-beta-D-Galactopyranosyl-oxymethyl-4-sulfatomethyl-furan (GSF) inhibited completly the adhesion of murine B16F10-cells and human WM-115-cell to fibronectin at 40 mM and to fibrinogen at 20 mM or rather 10 mM. GSF decreased the activation of MMP-2 and inhibited the migration of WM-115 and WM-266-4 cells at 5 mM. A significant reduction of the number of cells at 5 mM can only be seen after 60 hours due to cyctotoxic effects. So cytotoxicity is not relevant for the shown assays. The complete inhibition of cell-adhesion as well as the inhibitory effect of GSF on MMP-2 activation and cell-migration suggests an interaction with one or more members of the integrin family. The current work shows the potential especially of charged saccharide mimics to suppress the metastatic properties of melanoma cells in vitro. Therefore the combinatorial synthesis and screening of further charged saccaride mimics based on the furan-core with comparable properties in lower concentrations seems to be promising.
