Doxycyclin-kontrollierte Genexpression im ROSA26-Lokus der Maus

Abstract

In transgenen Mausmodellen häufig beobachtet und unerwünscht sind unvorhersehbare Einflüsse regulatorischer Elemente des Insertionsortes auf das Expressionsverhalten der ins Genom eingebrachten Gene. In der vorliegenden Arbeit wurde untersucht, ob diese Einflüsse durch Integration in einen definierten Genlokus kontrolliert bzw. minimiert werden können. Hierfür wurde der ROSA26-Lokus ausgewählt, für den gezeigt war, daß er in der Maus ubiquitär exprimiert und für Gen-Targeting leicht zugänglich ist. Zudem führt die Inaktivierung der ROSA26-Transkripte zu keinem offensichtlich veränderten Phänotyp. Die Induzierbarkeit von Fremdgenen im ROSA26-Lokus wurde durch das Einbringen von zwei unterschiedlichen, Tet-induzierbaren Modulen zur Expression von GFP, getestet. Nach homologer Rekombination werden der Tetracyclin-abhängige Transaktivator (itTA) bzw. der reverse Tetracyclin-abhängige Transaktivator (iM2) konstitutiv unter Kontrolle des endogenen ROSA26-Promotors exprimiert. In ES-Zellen konnte für das iM2-System eine Doxycyclin-abhängige Induktion von GFP und für das itTA-System eine Repression der GFP-Expression gezeigt werden. In heterozygoten iM2-Mäusen konnte, nach Deletion der Selektionskassette, in allen analysierten peripheren Geweben eine Induktion der GFP-Expression erreicht werden, wobei die GFP-Expressionsniveaus gewebsspezifisch variierten. Induktionszeitpunkt und Dauer zeigten keinen Einfluss auf die GFP-Expression. In homozygoten iM2-Mäusen wurde eine gewebsabhängige Steigerung der GFP-Expression beobachtet. Im Gehirn heterozygoter iM2-Mäusen konnte keine, und in homozygoten iM2-Mäusen nur eine auf definierte neuronale Populationen beschränkte Induktion der GFP-Expression erreicht werden. Transaktivierungsexperimente, sowie inhomogene GFP-Expression im Gehirn heterozygoter itTA-Mäuse indizieren ein Silencing des ROSA26-Lokus in Neuronen, was sich in einer schwachen Aktivität des ROSA26-Promotors sowie einer schlechten Induzierbarkeit des CMVm auswirken kann. In dieser Arbeit wurde gezeigt, dass die verwendeten Expressionsmodule zur Charakterisierung von Genloci und deren endogenen Einflüsse auf Fremdgene geeignet sind. Hierbei wurde deutlich, daß der ROSA26-Lokus aufgrund seiner gewebsspezifischen Expressionsniveaus nur bedingt für die Verwendung über Gen-Targeting integrierter induzierbarer Module verwendbar ist.

Translation of abstract (English)

A major drawback of traditional transgenic technologies is that positioning effects due to the integration-site can result in ectopic expression of the transgene. In this study I have analyzed to what extent these positioning effects can be controlled or minimized by integration of genes into a defined locus. For this the ROSA26-locus was chosen, which was well known for easy accessibility for gene targeting and its ubiquitous expression. Furthermore, inactivation of its gene transcripts produce no obviously altered phenotype. Inducibility of transgenes targeted into the ROSA26-locus was investigated by integration of two different tet-dependent GFP expression systems. Upon homologous recombination either the tetracycline dependent transactivator (htTA) or the reverse tetracycline dependent transactivator (hM2) were constitutively expressed under control of the endogenous ROSA26-promoter. Functionality of the systems was shown in ES cells by doxycyclin dependent induction of GFP-expression for hM2 and repression for htTA. After deletion of the selection cassette, hM2-mice show GFP-expression in all peripheral tissues analyzed, with a tissue dependent variation in expression levels. Time point and duration of induction were found to have no influence on induction of GFP-expression. In homozygous hM2-mice, a tissue dependent increase of GFP-expression was observed. However, induction of GFP-expression in the brains of heterozygous hM2-mice failed and was restricted to defined neuronal populations only in homozygous mice. Transactivation experiments as well as poor GFP-expression in neurons of htTA-mice indicate a silencing of the ROSA26 promoter in neurons, which might lead to reduced promoter strength or poor activation of the inducible CMVm. In this work we could demonstrate that the employed expression-systems are useful tools to characterize gene loci and influences of their endogenous regulatory elements on transgene expression. It became obvious that the ROSA26-locus due to its tissue dependent variation of expression levels has limited suitability for inducible systems targeted into this locus.
