Abstract

In der vorliegenden Arbeit wurde eine hochauflösende optische Messapparatur (CMLTT) entwickelt und aufgebaut, die auf einem konfokalen Multiphotonen-Laserscanning-Mikroskop basiert und eine optische Pinzette, ein Modul für innere Totalreflektionsmikroskopie (TIRFM), eine zusätzliche stationäre Multiphotoneneinkopplung sowie Positionsdetektoren enthält. Dabei wurde diese CMLTT-Gesamtapparatur so konzipiert, dass die einzelnen Module weitgehend parallel einsetzbar sind. Der Laserstrahl des Laser tweezers- (optische Pinzette-) Moduls ist mit Hilfe von akustooptischen und elektrooptischen Modulatoren sowohl räumlich wie zeitlich modulierbar. Das TIRFM-Modul ist für zwei Anregungslichtquellen ausgelegt, die Leistung der stationären Multiphotoneinkopplung kann variiert werden. Für die Laser tweezers-Anwendungen stehen insgesamt drei Vierquadranten-Positionsdetektoren zur Verfügung, so dass eine Positionsmessung eingefangener Objekte in allen Mikroskopiemodi möglich ist. Diese modulare CMLTT-Messapparatur wurde an Myofibrillenpräparaten und Einzelmolekülsassays getestet. Sie erlaubte erstmals, hochauflösende Second Harmonic Generation Messungen der 3D-Sarkomerstruktur von Muskelzellen durchzuführen. Darüber hinaus gelang es, die Multiphotonenfreisetzung des Kalziumkäfigmoleküls DMNPE-4 zu realisieren. Auf Einzelmolekülebene wurde die CMLTT-Apparatur benutzt, um die kalziumaktivierte Motilität der molekularen Motoren Aktin und Myosin zu quantifizieren.

Translation of abstract (English)

In the present work, the development and implementation of a high-resolution optical measuring system (CMLTT) is described. This system is based on a confocal-multiphoton-laserscanning microscope and comprises an optical laser-tweezers module, total-internal-reflectance microscopy (TIRFM), an additional stationary multi-photon coupling and position detectors. The CMLTT system is designed so that each module can be used with others in parallel. The laser beam forming the optical tweezers can be modulated both in space and time using acoustooptical and electrooptical modulators. The TIRFM module is built to accustom two excitation light sources and the power coupling of the stationary multi photon module can be varied. For laser tweezers applications, three position detectors are available for all microscopic modes. This modular CMLTT microscopic apparatus has been tested on myofibrillar preparations and single molecule assays. For the first time, it was possible to carry out high resolution second harmonic measurements of the three dimensional sarcomeric structure of muscle cells. Additionally, the multi photon photo release of DMNPE-4, a calcium cage molecule, has been realized. Furthermore, the CMLTT system has been used to quantify the calcium activated motility of the molecular motor proteins actin and myosin.
