Abstract

Die Abhängigkeit des Nährstofftransports vom Wassertransport auf dem Weg durch das gleiche Transportsystem war die zentrale Frage dieser Arbeit. Um die Untersuchung dieser zentralen Fragestellung der Pflanzenernährung an intakten Pflanzen zu ermöglichen, wurde die Wurzeldruckkammern nach Passioura in wesentlichen Punkten fortentwickelt. Insbesondere wurde die Druckregelung deutlich flexibler gestaltet, auf ein Software-basiertes System umgestellt und die Hardware-Komponenten soweit als möglich minimiert. Zudem sollten Teilkomponenten des Transportsystems für Nährstoffe (Wurzelaufnahme, lateraler Transport auf der Transportstrecke) auf ihre Abhängigkeit vom Wasserfluss untersucht werden. Gemeinsam ist den untersuchten Nährstoffen Nitrat, Phosphat, Sulfat, Kalium, Calcium und Magnesium, dass der Massenfluss im Xylem aber auch die Aufnahme über die Wurzel stark reguliert ist und somit Änderungen des Transpirationsflusses ausgeglichen werden. Je nach Menge, chemischer Eigenschaft und physiologischer Funktion konnte der Massenfluss in Xylemstrom modifiziert werden. Vor dem Hintergrund von in der Literatur veröffentlichten Daten lassen sich die Ergebnisse dieser Arbeit wie folgt interpretieren. Während die Anionen-Massenflüsse dem Bedarf im Spross und dem Spross-Wurzel-Signaling entsprechend reguliert sind, kann der Kalium-Massenfluss durch seine massiven Xylem-Phloem-Relokation und seine Funktion als Ladungsausgleich variieren. Schwankungen im Magnesium-Massenfluss können entweder in der Vakuole oder in der Zellwandmatrix gepuffert werden. Bei Calcium wird auch ein kontrollierter Massenfluss sichtbar. Die Zellwandkapazität zur Calciumbindung kann als Puffer bei Massenflussvariationen angesehen werden. Der Nährstofftransport im Xylem der Pflanzen ist entsprechend als ionenspezifisch zu betrachten, wenn auch ein grundlegend über die Aufnahme regulierter Massenfluss bei allen Nährstoffen zu beobachten war. Da nur kurzfristige Abhängigkeiten im Stundenbereich untersucht wurden, können mittelfristige Transportprozesse mit diurnaler Rhythmik die Massenflüsse beeinflussen, ohne dass dies in den Experimenten sichtbar wurde. Der dargestellte experimentelle Aufbau ermöglicht zukünftige mittelfristige Untersuchungen des Nährstofftransports und seiner Abhängigkeit von internen Wasserflüssen unter kombiniert variierenden Transpirations- und Photosynthesebedingungen.

Translation of abstract (English)

Nutrient transport and it�s dependency from water transport using the same transport pathway were the main topics of this work. To enable the examination of this central question of plant nutrition using living plants, further developments of the root pressure chamber based on Passioura were necessary. Especially the pressure control was designed more flexible by software based realisation with minimised hardware components. In addition partial components of the plant transport system like nutrient uptake via the root or lateral ion exchange along the pathway were investigated according to their water flux dependencies. Common for all the nutrients like nitrate, phosphate, sulphate, potassium, calcium and magnesium is the strictly regulated ion mass flow in the xylem and the ion uptake by the root, balancing changes in transpiration fluxes. Depending on mass, amount, chemical properties and physiological functions the mass flow could be modified in the xylem stream. While mass flow of anions is regulated by sink demand and shoot to root signaling, there are good hints that potassium mass flow could vary because of it�s massive xylem phloem relocation and it�s function for charge balancing. Causes for calcium and magnesium mass flow variations were discussed. On the basis of these results the nutrient transport in the xylem of intact plants is regulated in an ion specific way. In addition a regulated uptake of all nutrients could be observed. This work focused on the influence of short term changing�s within hours in transpiration rate. With the developed set up the examination of mass flow processes influenced by diurnal rhythms will be possible in the future. The given set up will allow the analysis of nutrient transport and it�s dependencies from internal water fluxes under varied conditions of transpiration and photosynthesis.
