Abstract

TAFII250 („TBP associated factor 250“) ist die größte und funktional vielfältigste Unter-einheit des generellen Transkriptionsfaktors TFIID. Indem TAFII250 sowohl mit TBP, als auch mit nahezu allen TAFII-Untereinheiten interagiert, nimmt es eine zentrale Stellung inner-halb des TFIID-Komplexes ein. Zudem ist TAFII250 an der Erkennung der Initiator-Promotorsequenz beteiligt, kontaktiert Enhancer-gebundene Trankriptionsaktivatoren sowie verschiedene Komponenten der generellen Transkriptionsmaschinerie (GTM) und nimmt so eine zentrale Funktion bei der Core-Promotorerkennung und als Koaktivator ein. Insbesondere ist TAFII250 an der Aktivierung der Transkription von Zielgenen beteiligt, deren Genprodukte in essentielle biologische Prozesse, wie der Zellzyklusregulation und dem Zellwachstum, involviert sind. Ferner wurde gezeigt, daß TAFII250 über verschiedene enzy-matische Aktivitäten verfügt und neben einzelnen Komponenten der GTM auch Histone modifiziert. Im Rahmen dieser Arbeit konnte gezeigt werden, daß TAFII250 das Histon H2B phos-phoryliert. Dies läßt die Hypothese zu, daß die TAFII250-vermittelte Phosphorylierung von H2B an der Transkriptionsaktivierung beteiligt ist. Weitere Ziele der vorliegenden Arbeit waren, die H2B-spezifische Kinase-Aktivität funktional zu charakterisieren und Hinweise für ihre Beteiligung an der Transkriptionsaktivierung zu erhalten. Unter Verwendung verschiedener Histon H2B-Peptidmutanten konnte gezeigt werden, daß TAFII250 Serin 33 in H2B phosphoryliert. Dieses Ergebnis konnte mit Hilfe eines in dieser Arbeit generierten a-H2B- Phosphoserin-33-spezifischen Antikörpers bestätigt werden. Ferner konnten die Kinase-aktiven Domänen des TAFII250-Proteins auf zwei distinkte Bereiche eingegrenzt werden: eine NH2-terminale Kinase (NTK) und eine COOH-terminale Kinase (CTK). Beide Domänen sind sowohl auto- als auch transkatalytisch aktiv, wobei die Histonphosphorylierung von der CTK katalysiert wird. Des weiteren wurde gezeigt, daß etwa 600 der COOH-terminalen Aminosäuren der CTK für die Histonphosphorylierung essentiell sind. Durch computergestützte Analysen der TAFII250-Aminosäuresequenz konnten kata-lytisch aktive und insgesamt drei potentielle ATP-bindende Sequenzmotive innerhalb der NTK und CTK-Domänen identifiziert werden. Anhand dieser Ergebnisse sollte es möglich sein Kinase-inaktive Formen des TAFII250-Proteins herzustellen. Um einen Hinweis darauf zu erhalten, ob die CTK in vivo in den Prozeß der Transkriptions-aktivierung involviert ist, wurde mittels in situ-Hybridisierung die snail (sna)-Transkription in Ganzpräparaten von Drosophila Embryonen untersucht, die eine homozygot letale CTK-Deletionsmutante des TAFII250-Proteins enthalten. Die sna-Expression ist in den Embryonen deutlich reduziert, woraus abgeleitet werden kann, daß die CTK an der TAFII250- abhängigen Transkriptionsaktivierung beteiligt ist. Die Ergebnisse dieser Arbeit stellen somit einen Ausgangspunkt dar, die molekularen Mechanismen der TAFII250-abhängigen Transkriptionsaktivierung am Chromatin in vitro und in vivo zu entschlüsseln.

Translation of abstract (English)

TAFII250 („TBP associated factor 250“) is the largest and functional most diverse subunit of the general transcription factor TFIID. By interacting with TBP and almost all other TAFII-subunits, TAFII250 takes a central position within the TFIID-complex. Furthermore TAFII250 participates in the recognition of the initiator-sequence, contacts enhancer-bound transcription factors, as well as different components of the general transcription machinery (GTM). This way TAFII250 plays a central function in corepromotor recognition and as a coactivator. TAFII250 especially participates in the trancriptional activation of genes encoding for proteins involved in essential biological processes, like cell cycle control and cell growth. Further it has been shown, that TAFII250 possess different enzymatic activities and, in addition to single components of the GTM, also modifies histones. I could demonstrate that TAFII250 phosphorylates histone H2B. This observation supports the hypothesis, that TAFII250-mediated phosphorylation of H2B is involved in transcriptional activation. The intention of this work was to characterise the H2B-specific kinase-activity functionally and to determine whether the kinase-activity is potentially involved in transcriptional activation. By using different histone H2B-peptide mutants, it was possible to show, that TAFII250 phosphorylates serine 33 in H2B. This result could be confirmed by the use of a-H2B-Phosphoserin-33-specific antibodies generated in this work. Further the kinase-active domains of the TAFII250-protein were mapped to two distinct regions: a NH2-terminal kinase (NTK) and a COOH-terminal kinase (CTK). Both domains are auto- as well as transcatalytical active, whereby the histone-phosphorylation is mediated by the CTK. Furthermore I could show, that approximately 600 of the COOH-terminal amino acids are essential for the phosphorylation of histones. By computer-supported analysis of the TAFII250-amino acid sequence, catalytical-active and altogether three potential ATP-binding sequence motives within the NTK- and CTK-domains were identified. These results should make it possible to generate kinase-inactive forms of TAFII250. To see whether the CTK is involved in the process of transriptinal activation in vivo, the snail (sna)-transcription in whole mount embryos from Drosophila containing a homozygous lethal CTK-deletion mutant was determined by in situ-hybridization. The expression of sna in these embryos is dramatically reduces, indicating that the CTK is involved in TAFII250-dependent activation of transcription. The results of this work therefore represent a starting point to decipher the molecular mechanisms underlying the TAFII250-dependent activation of transcription on chromatin in vitro and in vivo.
