Abstract

In dieser Arbeit werden Probleme der Optimalen Versuchsplanung zur Parameterschätzung bei nichtlinearen DAE-Modellen behandelt. Es wird eine allgemeine mathematische Problemformulierung hergeleitet, zu deren Lösung geeignete numerische Methoden bereitgestellt werden. Diese wurden in einem Softwarepaket implementiert. Wir betrachten dynamische Prozesse, die durch Systeme Differentiell-Algebraischer Gleichungen modelliert werden können. Exemplarisch untersuchen wir Anwendungen für chemische Reaktionssysteme. Zur Modellvalidierung wird durch nichtlineare Parameterschätzung das Prozeßmodell an experimentelle Daten angepaßt. Die Signifikanz der Schätzung beschreiben wir mittels Sensitivitätsanalyse durch die Kovarianzmatrix. Zur Minimierung von Gütekriterien auf der Kovarianzmatrix formulieren wir Nichtlineare Optimale Versuchsplanungsprobleme. Optimierungsvariablen sind die Prozeßsteuerungen und das Meßlayout. Es handelt sich dabei um beschränkte Optimalsteuerungsprobleme. Zur Lösung setzen wir SQP-Verfahren ein. Die Zielfunktion hängt von Sensitivitäten der Modellfunktionen nach den Parametern ab. Wir benutzen Matrixableitungskalkül und Interne Numerische Differentiation in Verbindung mit Automatischer Differentiation, um alle benötigten Ableitungen effizient bereitzustellen. Wir formulieren Mehrfachexperimentprobleme, dabei können wir die Informationen aus Vorexperimenten mitberücksichtigen. Für die Behandlung der ganzzahligen Variablen zur Modellierung des Meßlayouts geben wir geeignete Relaxierungen und Heuristiken an. Da nichtlineare optimale Versuchspläne von der Unsicherheit der Modellparameter abhängen, untersuchen wir Ansätze zur Robusten Versuchsplanung. Zur Behandlung von allgemeinen Aufgaben dieser Problemklasse haben wir das Softwarepaket VPLAN entwickelt. Dessen Anwendung auf ausgewählte Praxisbeispiele aus der chemischen Reaktionskinetik zeigt, daß die Methodik erfolgreich zur Planung effizienter und effektiver Experimente eingesetzt werden kann.

Translation of abstract (English)

In this thesis we treat problems of optimum experimental design for parameter estimation in nonlinear DAE models. We develop a general mathematical problem formulation. For the solution, we provide suited numerical methods. They have been implemented in a software package. We consider dynamical processes, which can be modeled as systems of differential algebraic equations. As examples, we study applications to chemical reaction systems. To validate the models, we fit the process model to experimental data using the method of nonlinear parameter estimation. We describe the significance of the estimation by approximations of the confidence regions and the variance covariance matrix using sensitivity analysis. To minimize objective functions on the variance covariance matrix, we formulate nonlinear optimum experimental design problems. Variables for the optimization are process controls and the layout of the measurements. We obtain constrained optimization problems. To solve them, we employ SQP methods. The objective function depends on sensitivities of the model functions with respect to the parameters. We use matrix calculus and internal numerical differentiation together with automatic differentiation in order to provide all required derivatives efficiently. We formulate multiple experiment problems. Here we can take into account the a priori information from previous experiments. To treat the integer variables in the modeling of the experimental layout, we develop suited relaxations and heuristics. Because nonlinear experimental designs depend on the uncertainty of the model parameters, we investigate some approaches of robust experimental design. To solve general problems of this class, we have developed the software package VPLAN. Applying it to four selected practical examples from chemical reaction kinetics, we can show that the methods can be employed successfully in order to design efficient and effective experiments.
