Abstract

Die spektroskopischen Eigenschaften des stabilen NAD-Analogs carba-Nikotinamidadenindinukleotid (cNAD) wurden sowohl von physikalischer Seite als auch im Hinblick auf die Anwendbarkeit in einem optischen Glucosesensor untersucht. Entgegen der Literatur weist cNADH den gleichen Extinktionskoeffzienten e=6,2(2)mM^-1cm^-1 wie NADH auf. Die experimentell beobachtete Rotverschiebung des Absorptionsmaximums von 340nm (NADH) zu 360nm (cNADH) konnte auf Basis quantenchemischer Rechnungen nachvollzogen werden und begründet sich letztlich in der Ladungsverschiebung am Cyclopentanring. Die maximale Fluoreszenzemission tritt sowohl bei NADH als auch bei cNADH bei einer Wellenlänge von 464(2)nm auf, wobei cNADH mit Q=0,024(2) eine um 50% höhere Quantenausbeute als NADH besitzt. Es konnte ferner verifiziert werden, dass beide Fluorophore innerhalb der Fehlergrenzen die gleichen Fluoreszenzlebensdauern von t1=0,32(3)ns und t2=0,68(3)ns aufweisen. Mehrere Hypothesen zur Ursache des Auftretens von zwei Lebensdauern wurden untersucht. Fluoreszenzlöschung durch molekularen Sauerstoff, Protonierung /Deprotonierung im elektronisch angeregten Zustand und die cis/trans-Isomerie konnten ausgeschlossen werden. Ebenso wurde eine spektrale Abhängigkeit der Lebensdauern nicht beobachtet. Während ein 2-Zustands-Modell für NADH noch Fragen offen lässt, konnten die beiden Fluoreszenzlebensdauern von cNADH in H2O auf eine offene und eine geklappte Konformation des Moleküls zurückgeführt werden. Die Bindung des Coenzyms NADH bzw. cNADH an ein Enzym (hier: Glucosedehydrogenase) erhöht sowohl die Quantenausbeute als auch die Fluoreszenzlebensdauer (tgeb=2,9(1)ns). In diesem Kontext wurde die Fluoreszenzlebensdauermessung als neuartiges Verfahren zur zuverlässigen Bestimmung der Bindungskonstante zwischen Enzym und Fluorophor vorgeschlagen und experimentell demonstriert. Ferner wurde die Option der Fluoreszenzlebensdauermessung von cNADH für einen - auch bei variierender Enzymaktivität - robusten Glucosesensor belegt.

Translation of abstract (English)

The spectroscopic properties of the stable NAD-analogue carba-nicotinamide adenine dinucleotide (cNAD) were analyzed both from a physical point of view and regarding its applicability for glucose sensing. In contrast to literature the extinction coeffcient of cNADH and NADH match (e=6.2(2)mM^-1cm^-1). Quantum chemical calculations supported the redshift observed experimentally between the absorption maximum of cNADH (360nm) compared to that of NADH (340nm) and linked it to the charge transfer at the cyclopentane ring. The maximum of fluorescence emission occurs at 464(2)nm for both, NADH and cNADH, whereby the quantum yield of cNADH Q=0.024(2) is 50% higher compared to NADH. Furthermore, both cNADH and NADH show two fluorescence lifetimes (t1=0.32(3)ns and t2=0.68(3)ns) of identical values (within the error margins). Different hypotheses concerning the origin of the occurrence of two fluorescence lifetimes were investigated. Fluorescence quenching due to molecular oxygen, an excited state protonation/deprotonation as well as cis/trans isomerization could be excluded. Moreover, the fluorescence lifetimes show no spectral dependence. While a two-state model for NADH leaves some questions unanswered, the two lifetimes of cNADH in H2O could be related to an open and a folded conformation of the molecule. If the coenzymes NADH or cNADH bind to an enzyme (here glucose dehydrogenase) the quantum yield as well as the fluorescence lifetime increases (tgeb=2.9(1)ns). In this context the use of fluorescence lifetime detection was proposed and successfully demonstrated as a novel approach for the reliable determination of the association constant between an enzyme and a fluorophore. Furthermore, it was shown that the fluorescence lifetime measurement of cNADH can be applied to glucose sensing even at varying enzyme activity.
