Abstract

In dieser Arbeit wurde ein Verfahren zum Nachweis von modifizierten Nukleotiden (DNA-Addukten) basierend auf Fluoreszenzderivatisierung und kapillarelektrophoretischer Trennung mit Laser-induzierter Fluoreszenzdetektion entwickelt. Es wurde eine Derivatisierungsstrategie gefunden, die eine Kopplung von 2'-Desoxynukleosid- 3'-phosphaten aus enzymatischer DNA-Hydrolyse mit Fluoreszenzmarkern, die eine primäre Aminogruppe als Ankergruppe tragen, ohne Zwischenreinigung ermöglicht. Zur Trennung fluoreszenzmarkierter Nukleotide erwies sich die micellare elektro- kinetische Chromatographie in unbeschichteten fused-silica-Kapillaren und SDS als Micellenbildner als geeignet. Die entwickelte Methodik wurde zum Nachweis von 8-Oxo-dG, etheno-dA und 5-Methyl-dC aus definiert modifizierten Oligonukleotiden angewendet. In Kalbsthymus-DNA und DNA isoliert aus humanem Gewebe wurde 5-Methyl-dC direkt nachgewiesen und quantifiziert. Anhand der Analyse von mit Aristolochiasäure I in vitro-modifizierter Kalbsthymus-DNA gelang der simultane Nachweis unterschiedlicher DNA-Addukt-Klassen. Die entwickelte Methodik ermöglicht die Verwendung von DNA-Addukten als Biomarker genotoxischer Substanzen in molekular-epidemiologischen Studien.

Translation of abstract (English)

This work describes the development of an analytical method for the detection of DNA adducts based on fluorescence labeling and separation via capillary electrophoresis. A labeling method was developed for the coupling of a fluorescent dye with a primary aminofunction to the 3'-phosphate-group of deoxynucleotides, obtained by enzymatical digestion of DNA. A capillary electrophoretic separation was developed using SDS as micelles and uncoated fused-silica capillaries. The method was used for the detection of 8-Oxo-dG, etheno-dA and 5-Methyl-dC in modified oligonucleotides. 5-Methyl-dC was also detected and quantified in calf thymus DNA and DNA isolated from human tissue. The analysis of in vitro incubated calf thymus DNA with aristolochic acid I showed simutanous detection of alkylated and bulky DNA modifications. The method described enables the use of DNA adducts as bioamrkers for genotoxic compounds in molecular-epidemiologic studies.
